The ciliated olfactory epithelium in this richly vascularized space makes it ideal for rapid absorption into the systemic circulation, potentially leading to a faster response than oral medications. Potential benefits of delivery to this space also include the reduction of drug-related adverse effects and potential avoidance of delayed absorption in the GI tract and first-pass metabolism in the liver.
Traditionally, nasal drug administration has focused on delivery to the lower nasal space, which is associated with significant drug loss from the nose with existing delivery systems, which may lead to variability in absorption. In addition, deep inspiration associated with existing systems can collapse the nasal valve essentially blocking access to the vascular rich upper nasal space.
The mucosal layer in the upper nasal space tends to be more permeable and stable than the lower nasal cavity, permitting consistent, faster and more extensive absorption of drug into the underlying vasculature with less mucociliary clearance.

Results were from a poststudy questionnaire, which asked 354 people with migraine to rate their experience with Trudhesa against their best usual care. The results were patient-reported and not validated.

Other things you need to know


  • Before taking Trudhesa, talk to your healthcare provider (HCP) about all the medications you are taking
  • Do not take Trudhesa if you are pregnant, planning to become pregnant, or nursing
  • Before taking Trudhesa, talk to your HCP if you have, or think you may have, any cardiovascular (CV) disease risks
  • Trudhesa was not associated with drowsiness
  • Trudhesa can be taken twice within a 24-hour period, if needed, and 3 times over the course of a week
  • Generally low levels of nausea were seen with Trudhesa

People with migraine were excluded if they had a history of CV events or presented with significant risk factors for CV disease. But people with a history of hypertension, if stable and well controlled on current therapies for >6 months, could enroll if no other risks were present.
